Molecular-cytogenetic analysis of AML patient with complex karyotype:
a) mFISH result identified complex rearrangements of chromosomes 2, 3, 7, 10, 17, 20, 21 with apparent dicentric chromosome dic(9;11); b) mBAND method for chromosome 9 confirmed presence of whole chromosome 9 in a derivative chromosome der(9;11) with inserted duplication of short arm dup(9)(p24p23); c) FISH analysis with multi-centromeric probe proved α-satellite region of chromosome 11 on der(9;11) but any α-satellite region of chromosome 9 – the loss is evidence for stabilization mechanism of dicentric chromosome which resulted in secondary monocentric chromosome; d + e) FISH analysis with combination of centromeric probes for chromosomes 13, 21 (SpectrumGreen) and 14, 22 (SpectrumOrange) verified presence of α-satellite chromosome 21 region (without typical constriction form) on pseudodicentric derivative chromosome psu dic(10;21)(21pter→21q22.3::21q22.1→21q22.3::10p13→10qter).
