Laboratory of Recombinant Vaccines

The main interest lies in the development of protocols for ex vivo expansion of multi-specific T cell lines that could be used for the prevention of severe viral infections in immunosuppressed patients or as a substrate for the preparation of genetically modified T cell lines. T cells are stimulated with various forms of antigen such as dendritic cells electroporated with synthetic in vitro transcribed mRNA coding for viral proteins or mixtures of overlapping synthetic peptides derived from viral antigens and by cultivation in the presence of cytokines. In parallel, chimeric antibody receptors (CARs) with specificity for virus encoded antigens or leukemia associated antigens for genetic modification of T cells are prepared in co-operation with the immunotherapeutic group.

Recently, we have introduced an assay for the detection of antigen specific cellular immunity against human cytomegalovirus (HCMV). Evaluation of this ELISPOT-interferon gamma test performed in a prospective study with 27 recipients of hematopoietic stem cells has revealed the suitability of the assay for the search for patients who are at high risk of reactivation of HCMV infection.
